Ambelline - hledání lepších zdrojů pro jeho izolaci a přípravu nových derivátů s cytotoxickým potenciálem

The proposed experimental project is focused on a very actual topic – the development of potential anticancer treatment. This project's main aim is to search for new and better natural sources of ambelline that can be used as a starting structure for semi-synthetic derivatives with significant cytotoxic activity. Our work will be oriented to screening different sources of ambelline to find out the most suitable one with the highest amount of this alkaloid. Ambelline is also available commercially, but the price is high, and there is only a limited number of distributors. Within the isolation of ambelline, we will obtain a broad spectrum of Amaryllidaceae alkaloids proper for further syntheses of alkaloidal analogs. Richer natural source of ambelline than we use right now will allow us to prepare a novel series of ethers derivated from the ambelline skeleton. We will characterize the structure vs. activity relationships of those originally designed substances and extend the tested substances portfolio. Furthermore, this project deals with screening those derivatives in terms of cytotoxicity and testing different tumor or leukemia cells. Compounds with significant activity will be examined in more detail to reveal their mechanism of action. We will also test the effect of these substances on the lines normal and quiescent cells to investigate them' selectivity. This experimental project is at the interface of the chemical, biological, and medical sciences.
