Vývoj liquisolid systému k cílené lokální terapii zánětlivých onemocnění tlustého střeva
Inflammatory bowel diseases are chronic inflammatory conditions with no effective cure and, moreover, most orally administered drugs used in therapy produce undesirable systemic side effects. Targeting the drug to the site of inflammation in the colon is therefore a suitable strategy to achieve long lasting remission. The pharmaceutical challenges in targeting orally administered drugs to the colon arise from the high variability in the conditions along the gastrointestinal tract in terms of pH, motility, viscosity and role of gut microflora; so a combination of several formulation approaches in designing an effective drug delivery system is necessary. The aim of this project is to formulate an oral colon-targeted drug delivery system of calcineurin inhibitor Cyclosporine A (CyA) for the local treatment of inflammatory bowel diseases (IBD). The novel approach of preparing liquisolid systems in combination with mucoadhesive bacterial degradable polymers and pH-dependent coatings will be employed to control drug release from the specific minitablets. The properties of the excipients and their combination as well as the suitable formulation will be assessed. A novel carrier system targeted into the colon for a more effective treatment of IBD with the lower required dose and minimized systemic side effects is the main target.
