PKPD modelování distribuce rifampicinu s prostorovým rozlišením a možné klinické důsledky.

The project’s aim is to contribute to the development and investigate the clinical application of a pharmacokinetic/dynamic model of Rifampicin distribution profiles in human hepatocytes in the context of TBC-treatment. Whereas standard approaches aim at giving time-profiles only, the proposed model incorporates spatial resolution as well. At the costs of more demanding mathematical and computer modeling, this is expected to yield more precise and localized information which may be crucial for pharmacotherapy in clinical practice, for instance through detecting toxic drug levels in the human liver. This interdisciplinary project (Pharmacy and Mathematics) is closely tied with GAUK project 1206517,,Mixed PDE/ODE modeling of drug distribution profiles for efficient pharmacotherapy”, where the emphasis is on the formulation of an appropriate mathematical model based on partial differential equations. The current project focuses on clinical questions and on computer implementation issues like parameter estimation, sensitivity analysis and model validation, but contribution to refinement of the mathematical model is not excluded.
