Tvorba nového typu biokompatibilních hemodialyzačních membrán k separaci biomolekul

End-stage renal disease (ESRD) is a life-threatening condition requiring regular hemodialysis. The inability of the kidney to remove uremic toxins leads to its accumulation in blood with consequence being severe complications that increase morbidity/mortality of these patients. Moreover, ESRD patients undergoing hemodialysis present chronic inflammation and systemic oxidative stress. Hemodialysis procedure per se was shown to induce neutrophil activation, leading to oxidative stress, therefore amplifying the inflammatory process, resulting in disease progression. Although, nowadays used hemodialysis membranes are highly biocompatible, long-term intradialytic contact of blood with artificial materials leads to neutrophil activation. To minimize the negative impact of hemodialysis and to improve solutes removal, highly porous silica thin film with accurate defined pore size obtained by sol-gel technique will be structured by endotemplate from self-assembly liquid crystals. The membrane surface will be further modified by coating with antioxidants. To evaluate the efficiency of the new membrane a lab-scale flow system enabling closed-loop circulation of blood will be settled for in vitro evaluation. The biocompatibility of the membrane will be tested though the changes in markers of inflammation, neutrophil activation and changes in erythrocytes in blood from healthy individuals. The membrane potential to diminish inflammatory and oxidative processes will be evaluated using blood from hemodialysis patients.
