Targeting the resistance in cancer therapy by inhibition of selected human carbonyl reducing enzymes.
The goal of the project is to test the effect of several protein kinase inhibitors (PKi) on the activity of human reductase from the aldo-keto reductase superfamily. AKRs play an important role in the deactivation of some cytostatics, e.g. anthracyclines, thereby reducing the effectiveness of the treatment and, consequently, resistance. Selective inhibition of these reductases leads to preservation of the expected cytostatic effect of the drugs and thus also to the suppression of cell resistance to the given treatment. Experimental approaches will include in vitro incubations with recombinant enzymes and work with tumor cell lines.
